The Developing of Biliary Liver Cirrhosis(fibrosis) in Rats by the Operation of BDL/S and the Application of Sodium Diatrizoate

Abstract
The studies of the establishment of potential liver fibrosis(cirrhoeis) were carried out in female Sprague-Dawley rate. Two groups (normal group and liver fibrosis induced group) of rate were prepared for developing of biliaiy liver fibrosis. For the development of liver fibrosis, rats were operated bile duct Hgation/seission(BDI/S). And sodium diatrizoate (16§·/§¸) were added to the rats daily for six weeks. After operation, rate were observed for 6 weeks and sacrified. Clinical biochemistry, N-terminal pre. collagen type III peptide (PNIIIP as marker of collagen synthesis) in sera and total collagen (as parameter of collagen accumulation) in a piece of liver tissue were investigated, and liver tissue was also stained for histological finding by H&E and trichrome(Masson).
The value of total collagen in liver tissue in six weeks after BDL/S operation and administration of sodium diatrizoate was 1.5¡­3 times higher than that of in 6 weeks after sham operation. The value of PNIIIP in sera was also significantly increased by 68.8% at one week after operation and 129¡­146% at two-six weeks after operation. However, no significant correlation between the total collagen and PNIIIP(r=0.89). Enzyme activities in sera (ALT, AST, ALP) were increased by 30¡­350% at one-two week after operation. But the enzyme activities were decreased in 6 weeks after operation. The amount of total bilirubin as parameter of jaundice was observed as high value for observation period. The collagen fiber was appeared at the surround of portal triad (portal vein, portal artery, bile duct) in sham operated rats.. Meanwhile, the thick collagen fiber and many collagen bundles at portal field and stroma in liver for BDLlB and sodium distrizoate treated groups were observed. The hepatocytes regeneration, bile duct proliferation and inflammation cell were shown stone week after operation. Massive bile duct proliferation and scar formation between bile duct and new formed bile duct were started at 2 weeks after operation. Hepatocytes in new formed bile ducts and micro nodules with portal-portal septum from massive proliferation of bile duct were isolated at three-sis weeks after operation.
All data represents that liver cirrhosis(fibrosis) was developed by BDL/S operation and sodium diatrizoate application. And the collagen synthesis was stimulated by these operation and carcinogenesis agent. The collagen metabolism, synthesis and accumulation can be monitored by measurement of PNIIIP in sera and total collagen in liver tissue.
